FITC-labeled AEDG, EDR, and KEDG entered HeLa cytoplasm, nuclei, and nucleoli, and fluorescence quenching suggested sequence- and methylation-sensitive DNA contacts — molecular work, not a fertility trial of Testagen (KEDG).

Methods

HeLa cells were incubated with fluorescein isothiocyanate–labeled short peptides: epithalon (Ala-Glu-Asp-Gly / Epitalon), pinealon (Glu-Asp-Arg), and testagen (Lys-Glu-Asp-Gly). The authors also measured fluorescence of 5,6-carboxyfluorescein–labeled deoxyribooligonucleotides and DNA–ethidium bromide complexes in the presence of intact peptides, including bronchogen (Ala-Glu-Asp-Leu), and report Stern–Volmer quenching constants. Findings

This paper is mechanistic or modeling evidence, not a clinical trial. The authors report marked fluorescence in cytoplasm, nucleus, and nucleolus after incubation with each labeled peptide, which they read as nuclear entry. Quenching constants differed by peptide sequence and by single- versus double-stranded labeled oligonucleotides, including sensitivity to cytosine methylation. They suggest epithalon, pinealon, and bronchogen prefer CNG-containing sequences, with epithalon, testagen, and pinealon preferring CAG and bronchogen CTG.

KEDG here is a molecular ligand in HeLa nuclei and on oligonucleotides. This is not a reproductive-organ trial and not evidence that Testagen treats fertility. Follow the PMID.
